He was born in Lancaster, Pennsylvania, three months after the death of his father. His mother returned to Philadelphia, after its evacuation by the British in the fall of 1778, and he was reared and educated in that city, spending his whole life there. He and his first wife were the parents of nine children of whom George Mifflin was the second.
